Butylene Oxide: Possibly carcinogenic to humans. (IARC-2B)
Carcinogenicity Data:
Although there is no direct evidence that exposure of people to 1-Bromopropane adversely affects
reproduction or development, studies reviewed by the expert panel and more recent studies in rats
show that exposure to 1-Bromopropane can adversely affect reproduction and development. (7)
The critical reproductive toxicity study in animals was a two-generation, inhalation study. It showed
exposure of rats to 250 ppm or greater altered numerous reproductive endpoints in both females and
males. These included decreased prostate weight, decreased sperm motility and percent normal sperm
in males and decreased litter size, decreased numbers of implantation sites, increased ovarian follicular
cysts, and increased estrous cycle length in females. The panel determined that no adverse
reproductive effects were observed at an exposure concentration of 100 ppm. (7)
Another recent study showed that female rats exposed 8 hours per day to 200, 400, or 800 ppm 1-BP
for 7 to 12 weeks showed a significant, dose-dependent increase in the number of irregular estrous
cycles, with extended diestrus at 400 ppm (Yamada et al., 2003). Additionally, examination of the
ovaries showed a dose-dependent reduction in the number of normal antral (or mature) follicles at
doses of 200 and 400 ppm. This effect is consistent with reduced fertility and litter sizes reported in
earlier studies. Interestingly, the number of primordial follicles was not significantly reduced in any of
the 1-Bromopropane exposure groups. The authors suggest that antral follicle counts may provide a
more sensitive indicator of 1-Bromopropane-induced female reproductive dysfunction than other
endpoints such as estrous length. (7)
Reproductive Data:
This product produced negative results for mutagenicity (Ames Test). (3)
Mutagenicity Data:
No adverse teratogenic effects are anticipated.
Teratogenicity Data:
None known.
Respiratory / Skin Sensitization
Data:
None known.
Synergistic Materials:
Butylene Oxide: Cancers of the respiratory system have been observed in rats and mice exposed by
inhalation. Negative results have been obtained in mice exposed by skin application. The International
Agency for Research on Cancer (IARC) has concluded that this chemical is possibly carcinogenic to
humans (Group 2B). (4)
When n-Propyl Bromide was tested for genetic toxicity, it tested negative for dominant lethal activity in
rats at 400 mg/Kg/day. This dose was given to male rats prior to breeding once weekly for eight
successive weeks. No difference in mating performance was observed. (9)
The frequency of fertile matings, mean number of corpora lutea, number of implants per female,
number of live embryos per female and the dominant lethal index was comparable to the negative
control group at weeks 1 through 8 after treatment. The frequency of dead implants was higher during
week eight compared to the controls, but no increases in dominant lethal index were observed during
the previous seven weeks. The frequency of dead implants in the treated group was similar to that of
the controls during weeks one through seven. (9)
